The association between obesity and related health problems has been well documented, yet little is known about the relationship between obesity and the disease itself.
In this prospective, randomized, double-blind, controlled study, we aimed to determine the incidence of obesity and the risk of diabetes mellitus associated with obesity. We included adult patients with type 2 diabetes mellitus treated with the following regimens: (1) a single dose of orlistat or placebo plus a metformin, (2) a single dose of orlistat plus metformin and a combination of orlistat plus metformin, (3) a single dose of orlistat plus metformin plus metformin, or (4) a single dose of orlistat plus metformin plus metformin. All patients were given a single dose of orlistat (orlistat 50 mg plus metformin) or a single dose of orlistat plus metformin (orlistat 120 mg plus metformin) for 12 weeks. The study was double-blinded. The primary endpoints were the primary outcome measure (serum glycosylated hemoglobin (HbA1c) levels) and the secondary outcome measures (serum triglycerides, total cholesterol, and fasting and postprandial serum lipids).
Data on primary outcome measures were analyzed using logistic regression. The primary endpoints were the secondary endpoints, and the risk of diabetes-related events was assessed using Kaplan-Meier survival analysis.
